The effects of Eucommial cortex and leaf extracts on the Growth rate of animal leukemic cells, L_(1210)


Abstract
This study was devised to observe the cytotoxic activity of Eucommial cortex and leaf extracts against the leukemic cell, Lazio, in vitro.
Each extract was prepared by extraction with water, 50%, and 95% ethyl alcohol, acetone, chloroform and petroleum ether, and then the cytotoxic activity of each extract was compared with control group and each other.
The effects of incubation time, size distribution of cell, cloning efficiency, and development of resistance in the culture medium against the growth of L1210 were determined after treatment of chloroform extract which had showed strongest activity.
The experimental results obtained are summarized as follows;
1. The cytotoxic activity of Eucommial cortex and leaf was strongest in the chloroform extract and that of water extract was weakest.
2. The specific activity of chloroform extract of Eucommial leaf was stronger than that of Eucommial cortex extract.
3. The cytotoxic activity of Eucommial cortex extract showed the highest value within 12 hours during incubation, but the activity of Eucommial leaf extract increased gradually in course of incubation time.
4. When the L1210 cells were incubated with the extracts of Eucommial cortex and leaf for 24 hours, the peak of size distribution of cell was shifted to direction of the small size region.
¢¥5. When L1210 cells were incubated in media containing 3 units or 1.5 units of the extract of Eucommial cortex or leaf, viable cells estimated by the soft agar cloning method were not present.
